How to use the nanodrop to measure DNA concentration | Biology Tutorial

 Overview 

After you purified your DNA it is essential to know the DNA concentration. An easy way to obtain that information is to do a Nanodrop measurement. It will quantify your DNA concentration within seconds and deliver reliable results. A big advantage is the low amount of sample you lose during measurement (1 µl).

Chemicals and Materials you will need: 

Protocol: 

  • start the nanodrop program
  • blank the machine 
    • clean the area of measurement with ethanol and water using a laboratory swipe
    • add 1 µl of the solution in which your DNA is dissolved (elution buffer) onto the area of measurement 
    • press blank
  • measure your sample 
  • add 1 µl of your DNA sample onto the area of measurement 
  • save your results 
  • clean the area of measurement with ethanol and water
