 How to use a drigalski spatula  Prepare a 96% Ethanol solution   Place the drigalski spatula in the ethanol  Flame the spatula and let the ethanol burn (to sterilize)  Place the spatula on the agar plate and close the plate with the lid (to cool spatula) Add your bacteria  Disperse the bacteria with the spatula until the agar is dry  Place the plate with bacteria in the incubator at a desired temperature  Place the spatula back in ethanol and store at RT

 How to make a bacterial liquid culture  Preheat incubator to desired temperatur  Preheat media to desired temperatur Pipetet desired volume of medium into erlenmeyer flask   Add antibiotic if necessary  Pick a single colony with a pipette tip and drop it into the flask  Move flask to incubator  Incubate over night or for a certain time period  Proceed to use bacteria for downstreamm application

How to refill a liquid nitrogen tank  The tank which is described in this video is a liquid nitrogen tank by Thermo Fisher Scientific .  Perform a manual refill before disconnecting the nitrogen supply Close the valves  Disconnect the nitrogen supply - no turning off is required  Move the tank to the nitrogen supply and connect the supply tubes  Open the valves slowly and let nitrogen in  Refilling takes 30 min to 1 hour   Reconnect the tank to the cell tank 

  Stripping of WB membranes protocol -          Prewarm stripping box for 10 min. in 55 °C water bath with 100 mL stripping buffer + 680  µ L  β -Mercaptoethanol (alternatively you can use 50 ml with 340 µl) -          Put membrane in stripping buffer box in water bath for 20 minutes, after incubation pour stripping buffer in  β -Mercaptoethanol waste -          Wash 1x with distilled water; add water and let it incubate without shaking to evaporate  β -Mercaptoethanol -          Transfer membranes into new box with TBST buffer -          Wash on shaker for 10 minutes 3x with TBST buffer  -          Prepare next antibody  -          Put in tubes with new antibody (no extra b...
